Dear Yogesh,
As for your enquiry regarding flat slabs with varying moments of inertia it can be done and has been done. Care has to be taken in modeling. The following article by Dr. Bijan Aalami, a renowned expert who developed the ADAPT software is a useful read.

Thanks sir,
I have another question.
Sir,
I am modeling flat slabs in staad. I have applied seismic forces. Staad shows some moments and shear for seismic forces. Accordingly I have designed the slab. I am considering the shear SQX and SQY at d/2 away from the face of the column. I am considering the global moments in X and Z direction. I am also considering MXY and add it to the moment to calculate steel. I am not designing the slab by IS method. I am providing the steel as per moment and stress.
My question is….
Shall I have to calculate additional moments due to unequal spans, additional shear while considering punching shear check? Is it necessary to satisfy the conditions given in IS456 regarding 3 nearly equal span and loading?
